Antiphospholipid Antibody Study
1) A CBC, serum biochemistry profile and urinalysis needs to be available for all cases
2) Case inclusion criteria = moderate anemia (PCV < 30) plus at least two of the following:
blood smear evidence of spherocytosis
positive saline agglutination test
positive coomb’s test
evidence of anti-erythrocyte antibody by flow cytometry
3) samples required
A. citrated plasma
i. fill a citrate containing vacutainer tube with whole
blood (blue topped tube)
ii. give to the clinical pathology laboratory ASAP
***SAMPLE SHOULD BE TAKEN BEFORE ANY HEPARIN ADMINISTRATION OR TRANSFUSION. PLEASE USE A NON HEPARIN FLUSH FOR CATHETER PLACEMENT.
B. serum
i. serum from biochemical profile may be used
ii. store at -70°C until shipping (in plastic tubes)
iii. at least 500 microliters are needed
iv. serum can be obtained post treatment
4) Shipping samples: Samples may be batched for ease of shipping.

citrated plasma for antiphospholipid study

“spin and save”
1) Centrifuge once at 5000 x g for 3 minutes
2) Harvest plasma and place in a plastic serum tube with a snap or screw cap
3) Save pellet with cellular elements in refrigerator
4) Centrifuge the collected plasma again at 5000 x g for 3 minutes
5) Collect the plasma and place in a plastic serum tube
6) Label the tube with the case number and date
7) Place in -70 freezer in the box labeled Dr. Olver’s aPL study
*The plasma needs to be separated from the cells and frozen within 4 hours of collection!
